Description

Description: This assay employs a two-site sandwich ELISA to quantitate MMP21 in samples. An antibody specific for MMP21 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP21 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP21 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP21 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Matrix metalloproteinase-21 is a member of the matrix metalloproteinase family. Proteins in this family are involved in the breakdown of extracellular matrix for both normal physiological processes, such as embryonic development, reproduction, and tissue remodeling,and disease processes, such as asthma and metastasis. The encoded protein may play an important role in embryogenesis, particularly in neuronal cells, as well as in lymphocyte development and survival.
Matrix metalloproteinases, such as MMP21, constitute a family of proteolytic enzymes that function extracellularly to degrade extracellular matrix proteins. Some MMPs also function along the secretory pathway and modulate the activities of growth factors or their receptors
